It gets quite underrated as an upset, but I think Junior Jones stretching an unbeaten Barrera was pretty shocking. Bojado getting beat by a journeyman was pretty big. And although in hindsight it shouldn't be considered such a big upset, Starling beating Mark Breland was a surprise at the time i've heard. I'm trying to think of some lesser known examples.
Honeyghan beating Curry is perhaps the best example in British boxing history of the underdog beating the favourite. Curry was something like P4P number 2 going into that fight and Honeyghan was an afterthought, yet Honeyghan beat him. Greatest achievement in British boxing history since Turpin beat Robinson I guess. I know it hasn't been surpassed since.
