As sad as it is to say, the three biggest defining moments of his career (in terms of specific fights) are his three losses. In due time, I think Cotto vs. Margarito I will be his most talked about, Cotto vs. Mayweather will be his most re-watched, and Cotto vs. Pacquiao will be a lesser combination of both.
If anything, the losses make Cotto even more endearing and admirable, IMO. Cotto handles himself like a true professional fighter. He has taken all of the challenges put in front of him. When you do that, inevitably you will likely lose a fight or two. Pac and May won't fight each other, but Cotto fought both. And the highly avoided (likely plastered up) Margarito. Edit: I know Margo wasn't undefeated.
Cotto is a great fighter, but the fight that sticks out to me was when he beat Judah. due to styles making fights, I thought Cotto could be in for real trouble and Judah did cause problems, but the way Cotto fought back and won was breath taking

I think his peak, and defining moment, was when he put on an awesome display of punching and boxing against a still dangerous mosley.
The Margarito revenge fight was his moment. The Judah fight was a solid encounter but the Mosley fight was too close to be a great victory. A rematch of that fight was in order at the time.

When he walked over to Margarito's corner after the second fight and stared him down. That was a great moment, and something I'll remember about Cotto for a long time. I haven't been that happy to see a boxer win a fight in a long time. That revenge was very well-deserved and a long time coming.
