Does a boxer's title become vacant when he moves between weight classes?

No, not automatically if that's what you mean.

It becomes vacant, only if he vacates or has been stripped of the title. A fighter can be a champion in a certain division and move up or down to challenge a title holder while still retaining their title in their respective weight class.

What if you move down? Do you have to put your belt on the line or call the fight an exhibition?

The recent SMW bout between GGG and Canelo should answer that question.
